Click the Start-All Programs-Accessories-System Tools-Disk Defragmenter window appears so that the Disk Defragmenter.
From Defragmenter window select the drive in which akan defrag, from the picture above there are two drives in which you want to defrag the drive C and drive D.
For the first time do the defrag on drive C that is the way click on Drive C and click Defragment, and wait until the defrag process has finished.
After the defrag process is complete it will appear a message that the defrag has finished.
If you have finished making defrag C adrive pad, then continue with the defrag the files on drive D.

Yes, the HDD being the slowest component in the data chain, plays an important role in the overall speed and must be maintained well. Its important to run diskcleanup and defragmemt regularly. Now its easy to defrag since programs like Diskeeper (the best among defraggers I have tried)can be set to run in the background in realtime automatic mode.
